conditions. The live transport of small-size juveniles to grown-out sites induces hypoxia and congesting stress. The transport process is essential to production of outdoor healthy seedlings. Here, we evaluated the effects of damp transport and water transport on Apostichopus japonicus, an economic species inhabiting along the Asian coast. The survival after transport and immune performance of small-size juveniles (0.05-0.1 g of immersed weight) were monitored during distinct duration of transport (1, 3, 5, 8 and 12 hr). The results revealed that there were no significant differences between the two transport systems within 5 hr. Given the survival for recovery of 48 hr and responses of immune enzymes and catecholamines during transport, water transport was less detrimental to sea cucumbers than damp transport when the transport duration was less than 8 hr. Once the duration prolonged to 12 hr, these immune parameters changed irregularly and the survival declined drastically. Water transport with a notably higher survival for juveniles was the more appropriate manner for long-time transport. We have found that MITF participates in the immune response to Vibrio parahaemolyticus infection in the clam Meretrix petechialis. In this study, we focused on how MITF functions in immunity. First, PO, CTSK, and BCL-2 were identified as the target genes of MpMITF in the clam by RNAi. EMSAs showed direct binding between the MpMITF protein and the E-box of the MpPO, MpCTSK, and MpBCL-2 promoters. Yeast one-hybrid assays also suggested that MpMITF could activate the expression of these three downstream genes. These results demonstrated that the transcriptional expression of MpPO, MpCTSK, and MpBCL-2 is directly regulated by MpMITF. Second, we analyzed the roles of MpPO, MpCTSK, and MpBCL-2 in clam immunity. The mRNA expression of MpPO, MpCTSK, and MpBCL-2 increased significantly after V. parahaemolyticus challenge, which implied that these genes might take part in the immune defense against V. parahaemolyticus challenge in clams. The purified recombinant proteins, MpPO and MpCTSK, inhibited the growth of V. parahaemolyticus. Additionally, the apoptosis rate of clam haemocytes rose significantly when the activity of MpBCL-2 was suppressed. These results revealed that MpPO, MpCTSK, and MpBCL-2 are involved in the immune defense against V. parahaemolyticus. Measurement of the diesel-oil degradation rates of these strains by the gravimetric method revealed that they ranged from 42% to 66% within 2 weeks. The highest degradation rates were observed from strains CQ8-1 (66%), CQ8-2 (62.6%), and CQ11 (59%), which were identified as Bacillus thuringiensis, Ochrobactrum anthropi, and Bordetella bronchialis, respectively, based on their 16S rDNA sequences. Moreover, the physiological and biochemical properties of these three strains were analyzed by Gram staining, catalase, oxidase, and Voges-Proskauer tests. Transmission electron microscopy showed that all three strains were rod shaped with flagella. Gas chromatography and mass spectrometric analyses indicated that medium- and long-chain n-alkanes in diesel oil (C11-C29) were degraded to different degrees by B. thuringiensis, O. anthropi, and B. bronchialis, and the degradation rates gradually decreased as the carbon numbers increased. This study integrated isotopic geochemical measurements with high-throughput sequencing of 16S rRNA gene amplicons to evaluate the source of groundwater salinity and the influence of hydrogeochemical variations on microbial communities under different salinity gradients in the Pearl River Delta of China. Results showed that the groundwater salinity in this area varied from fresh water in the inland area to brackish water, and then to saline water close to the southeast shoreline. The major ions (Na+, K+, Ca2+, Mg2+, Cl-, NO3-, SO42-, and HCO3-) and isotope analyses (H-2, H-3, O-18, and C-14) indicated that the groundwater in the confined aquifer was recharged by local precipitation and seawater. A further C-14 analysis showed that the salinity of the groundwater was likely attributed to the Holocene transgression. Analysis of the microbial community showed that -proteobacteria were frequently observed in all the groundwater samples, while the other main microbial community at class level varied greatly, from -proteobacteria in the freshwater wells to epsilon-proteobacteria in the brackish wells and to Bacilli in the saline wells. Exiguobacterium and Acinetobacter were dominant in saline water and the brackish water sample of Q144, while Sulfuricurvum dominated in the brackish water sample of Q143. Aeromonas, no rank Gallionellaceae, no rank Methylophilaceae, Acidovorax, and Comamonas unevenly thrived in the freshwater samples collected from different locations. The effect of temperature on the corrosion behavior of the 904L austenitic stainless steel in static seawater, which simulated the static period of seawater pumped storage systems, was determined by Tafel polarization and EIS. Based on the results of the electrochemical measurements, the corrosion rate of 904L ASS was found to increase with rising temperatures. 904L ASS has excellent corrosion resistance in static seawater.
